P-Selectin also known as CD62P or GMP-140 is a cell adhesion molecule expressed on the surface of activated endothelial cells and platelets. It has a molecular weight of approximately 140 kDa. In resting cells P-Selectin resides within alpha granules of platelets and Weibel-Palade bodies of endothelial cells. Upon activation it rapidly translocates to the cell surface playing an important role in mediating the initial interactions between endothelial cells leukocytes and platelets by binding to its ligand P-Selectin glycoprotein ligand-1 (PSGL-1).
P-Selectin facilitates the recruitment of leukocytes to the site of injury or inflammation. It is a member of the selectin family of cell adhesion molecules which also includes E-Selectin and L-Selectin. P-Selectin interactions promote rolling and tethering of leukocytes on the endothelium a critical step for leukocyte extravasation into tissues. These processes are important for innate immune responses and the inflammation process.
P-Selectin acts within the coagulation and inflammation pathways. In the coagulation pathway P-Selectin enhances platelet aggregation by interacting with fibrin and promoting thrombus formation. In the inflammation pathway it interacts with chemokines and integrins such as ICAM-1 facilitating leukocyte adhesion and transmigration. By partnering with these molecules P-Selectin coordinates cellular events that lead to immune response and wound healing.
P-Selectin is implicated in atherosclerosis and thrombosis. In atherosclerosis excessive expression of P-Selectin contributes to the recruitment of monocytes to the arterial wall promoting foam cell formation and plaque growth. In thrombosis elevated P-Selectin levels enhance platelet-leukocyte interactions exacerbating thrombus formation. The protein VWF (von Willebrand factor) is closely related to P-Selectin in these conditions as it also promotes platelet adhesion and aggregation.
