Purified ab76255 at 1/20 dilution (1μg) immunoprecipitating p130 in Jurkat was serum starved for 24h, then treated with 10% FBS for 20h whole cell lysate.

Lane 1 (input) : Jurkat (Human T cell leukemia T lymphocyte) was serum starved for 24h, then treated with 10% FBS for 20h whole cell lysate 10μg

Lane 2 (+) : ab76255 + Jurkat was serum starved for 24h, then treated with 10% FBS for 20h whole cell lysate.

Lane 3 (-) : Rabbit monoclonal IgG (ab172730) instead of ab76255 in Jurkat was serum starved for 24h, then treated with 10% FBS for 20h whole cell lysate.

VeriBlot for IP Detection Reagent (HRP) (ab131366) (1/1000 dilution) was used for Western blotting.

P130 also known as RBL2 (retinoblastoma-like 2) is a member of the retinoblastoma (Rb) protein family which includes pRb and p107. This protein has a molecular weight of about 130 kDa. It plays roles in regulating cell cycle by binding to E2F transcription factors consequently suppressing cell-cycle progression from G1 to S phase. p130 is ubiquitously expressed in various tissues but it shows higher levels in tissues with a low proliferation rate.
Biological function summary

The retinoblastoma-like 2 (p130) functions as an important regulator within the cell cycle checkpoint machinery. p130 forms a complex with E2F transcription factors selectively inhibiting their transcriptional activity. This inhibition plays an integral role in controlling cellular proliferation and ensuring that DNA replication does not occur unchecked. It also associates with cyclin-dependent kinase inhibitors further influencing the progression of the cell cycle.

Pathways

P130 is implicated in the control of the G1/S transition within the cell cycle pathway. Its function is closely linked with the Rb/E2F pathway which is critical for the regulation of cell proliferation. p130 collaborates with Rb and p107 within this pathway ensuring proper control of cell cycle transitions and maintenance of cellular growth.

Retinoblastoma-like 2 (p130) shows significant connections to cancer and neurodegenerative disorders. Directly involved in heterochromatin formation by maintaining overall chromatin structure and, in particular, that of constitutive heterochromatin by stabilizing histone methylation. Recruits and targets histone methyltransferases KMT5B and KMT5C, leading to epigenetic transcriptional repression. Controls histone H4 'Lys-20' trimethylation. Probably acts as a transcription repressor by recruiting chromatin-modifying enzymes to promoters. Potent inhibitor of E2F-mediated trans-activation, associates preferentially with E2F5. Binds to cyclins A and E. Binds to and may be involved in the transforming capacity of the adenovirus E1A protein. May act as a tumor suppressor.
