Supplementary information

The P2Y1 receptor also known as P2RY1 is a G protein-coupled receptor with a molecular mass of approximately 42 kDa. This receptor is expressed in various tissues including platelets endothelial cells and certain brain regions. P2Y1 primarily binds to extracellular nucleotides such as ADP and ATP initiating signaling pathways that involve intracellular calcium mobilization and activation of phospholipase C. Its role as a receptor for purines distinguishes it as a significant component in cellular communication processes.
Biological function summary

The P2Y1 receptor participates in platelet aggregation and the regulation of blood clot formation. It exists as part of a multi-receptor complex but can function independently. P2Y1 activation leads to shape change and aggregation of platelets contributing to hemostasis. Additionally its expression in neuronal tissue suggests involvement in neurotransmission and cognitive functions although the full spectrum of its biological roles continues to be studied.

Pathways

P2Y1 integrates into signal transduction cascades involving phosphoinositide metabolism and the regulation of intracellular calcium. The receptor is a part of the purinergic signaling pathway which includes other receptors like P2Y12. These signals play a role in platelet activation and vascular integrity where P2Y1 works in conjunction with other proteins like Gq alpha subunit to propagate cellular responses essential for maintaining vascular homeostasis and responding to vascular injury.

P2Y1 exhibits connections to thrombotic disorders and neurological conditions such as stroke. The receptor's role in platelet activation implicates it in the pathogenesis of thrombotic events where excessive or dysfunctional aggregation can lead to disorders like deep vein thrombosis. In the context of neural activity abnormal P2Y1 signaling may influence the risk or severity of ischemic stroke interacting with cyclooxygenase pathway components involved in inflammation and cerebral blood flow regulation. Understanding these interactions solidifies P2Y1 as a promising target for therapeutic interventions in these conditions.

Target data

Receptor for extracellular adenine nucleotides such as ADP (PubMed : 25822790, PubMed : 9038354, PubMed : 9442040). In platelets, binding to ADP leads to mobilization of intracellular calcium ions via activation of phospholipase C, a change in platelet shape, and ultimately platelet aggregation (PubMed : 9442040).
