Activity summary

P2Y4 also known as P2RY4 is a G protein-coupled receptor (GPCR) that responds to nucleotides like UTP and ATP. It has an approximate mass of around 39 kDa. This receptor is expressed in a variety of tissues such as the heart lung and intestine. Mechanically P2Y4 is involved in signaling pathways that activate phospholipase C leading to the release of intracellular calcium which plays a role in cellular communication and function.

Biological function summary

P2Y4 mediates responses essential for processes like gastrointestinal secretion and cardiovascular regulation. This receptor is part of the P2Y family of nucleotide receptors that influence diverse physiological actions through their complex interactions. It plays an important role in the regulation of ion transport and smooth muscle contraction by altering cellular cAMP and inositol trisphosphate levels through Gq/11 protein activation.

Pathways

The P2Y4 receptor plays a role in the purinergic signaling pathway and is also involved in the phosphoinositide signaling pathway. Within these pathways it interacts with other P2Y receptors such as P2Y2 and P2Y6 which share some functional and structural similarities. These pathways contribute to a variety of cellular responses including modulation of inflammation and tissue repair mechanisms.

Associated diseases and disorders

P2Y4 has associations with conditions like cystic fibrosis and certain cardiovascular diseases. In cystic fibrosis the disruption in the purinergic signaling pathway involving P2Y4 influences electrolyte and fluid balance affecting mucus viscosity. This receptor also interacts with other proteins such as CFTR and ENaC which are critical in epithelial ion transport abnormalities seen in these diseases. In cardiovascular diseases altered P2Y4 expression affects vascular tone and blood pressure regulation often linking it to related GPCRs involved in vascular health.
