Supplementary information

The p400 protein also known as EP400 plays a mechanical role as an ATPase in chromatin remodeling. Its molecular mass is approximately 420 kDa. This protein is predominantly expressed in tissues with high proliferative capacities such as testis and lymph nodes. It functions within the TIP60 complex also consisting of other important histone acetyltransferase components that regulate transcriptional activation and DNA repair processes.
Biological function summary

The p400 protein contributes significantly to gene expression and chromatin structure. It participates in the modification of chromatin architecture facilitating the access of transcription factors to DNA. p400 becomes an integral component of the TIP60 complex vital for the acetylation of nucleosomal histones which promotes transcriptional activation. Through these mechanisms p400 influences cellular processes such as cell cycle regulation and apoptosis.

Pathways

P400's functionality integrates into significant cellular pathways like the cell cycle and DNA repair pathways. Within the cell cycle p400's role in chromatin remodeling links it to the regulation of genes controlling cell proliferation. The protein interacts with histones and is part of complexes that include proteins such as TRRAP contributing to the DNA damage response and repair mechanisms. These connections highlight p400's involvement in cellular defense and maintenance of genomic stability.

P400's dysfunction associates with cancer and neurodegenerative disorders. Abnormal regulation of p400 due to its function in transcription and repair implicates its involvement in tumorigenesis particularly in cancers such as colorectal cancer. In neurological contexts aberrant p400 activity relates to neurodegenerative diseases like Alzheimer's where chromatin structure and gene expression go awry. The target’s interconnectedness with other proteins like TIP60 during the onset of these conditions highlights its importance in pathophysiological processes.

Target data

Component of the NuA4 histone acetyltransferase complex which is involved in transcriptional activation of select genes principally by acetylation of nucleosomal histones H4 and H2A. This modification may both alter nucleosome - DNA interactions and promote interaction of the modified histones with other proteins which positively regulate transcription. May be required for transcriptional activation of E2F1 and MYC target genes during cellular proliferation. The NuA4 complex ATPase and helicase activities seem to be, at least in part, contributed by the association of RUVBL1 and RUVBL2 with EP400. May regulate ZNF42 transcription activity. Component of a SWR1-like complex that specifically mediates the removal of histone H2A.Z/H2AZ1 from the nucleosome.
