May regulate the release of adrenocorticotropin, luteinizing hormone, growth hormone, prolactin, epinephrine, and catecholamine. May play a role in spermatogenesis and sperm motility. Causes smooth muscle relaxation and secretion in the gastrointestinal tract.

The PACAP receptor also known as ADCYAP1R1 is a G-protein-coupled receptor that interacts with the neuropeptide PACAP (Pituitary Adenylate Cyclase-Activating Polypeptide). This receptor plays a role in transmitting extracellular signals into cellular responses. It is comprised of approximately 525 amino acids with a considerable molecular mass of around 59 kDa. The PACAP receptors are widely expressed in the central nervous system including regions like the hypothalamus hippocampus and cerebellum. It also appears in peripheral tissues such as the adrenal glands and testis.
PACAP receptor plays significant roles in both neuronal and endocrine systems. It modulates neurotransmitter release and affects neurotrophic activities. The receptor often operates in a receptor-ligand complex with PACAP which enables it to participate in neuroprotective and neurite outgrowth functions. This receptor actively regulates activities within the hypothalamic-pituitary-adrenal axis impacting stress responses and circadian rhythms.
Many processes involve the PACAP receptor through essential signaling pathways like the cAMP-PKA and MAPK pathways. These pathways integrate signals from multiple receptor types including glucocorticoid receptors.
