PACE4 also known as PCSK6 is a member of the proprotein convertase family that functions mechanically as a serine endoprotease. This protein plays a role in cleaving precursors of hormones and other proteins to activate their bioactive forms. PACE4 has a molecular mass of approximately 97 kDa. It is expressed in various tissues including the brain pituitary gland and reproductive organs indicative of its involvement in diverse physiological processes.
PACE4 participates in cellular and developmental processes by processing precursors into active proteins. It is part of a complex of proprotein convertases responsible for the maturation of secretory pathway proteins. PACE4 takes part in the regulation of growth factors matrix metalloproteinases and adhesion molecules exerting influence on cellular growth and development as well as extracellular matrix remodeling.
PACE4 involves itself in specific signaling pathways integral to the activation of precursor proteins. It prominently participates in the TGF-beta signaling pathway where it activates transforming growth factor-beta precursors. PACE4 also acts in pathways implicated in the release of active forms of proteins such as integrins and collagens working alongside related convertases such as furin which shares overlapping substrate specificity.
PACE4 contribution links to pathological processes in certain cancers and osteoarthritis through its regulation of tumor growth and cartilage degradation respectively. In cancer PACE4 activates matrix metalloproteinases that aid tumor invasion and metastasis. Its relation with cancer involves interactions with other proteins like MMP-2 enhancing the potential for cellular invasiveness. In osteoarthritis PACE4's influence on cartilage tissue involves connections with ADAMTSs impacting cartilage matrix composition and health.
