PACS1 also known as PACSONE and PACS-1 is a cytosolic sorting protein that plays a role in maintaining intracellular cargo and membrane trafficking. It has a molecular weight of approximately 110 kDa. Expressed in various tissues including brain kidney and liver PACS1 works as a bridge connecting trans-Golgi network (TGN) to endosome trafficking pathways where it helps in targeting proteins for endocytosis and recycling back to the TGN.
Biological function summary

PACS1 interacts with various adaptor proteins to regulate the localization of critical signaling molecules within the cell. Its function involves linking to clathrin and interacting with other proteins to form stable complexes ensuring the proper sorting of cargo proteins. This sorting is essential for cell signaling and responses. PACS1's ability to interact with kinases and phosphatases helps regulate the phosphorylation state of other proteins impacting diverse cellular pathways.

Pathways

PACS1 is involved in key intracellular trafficking and protein modification pathways. In particular it plays a significant role in the endocytic recycling pathway which is important in recycling membrane receptors. It closely interacts with clathrin and adaptor protein complexes. Additionally PACS1 is related to the apoptosis pathway through its regulatory effects on protein kinase C. By influencing these pathways PACS1 helps maintain cellular homeostasis and modulate cell survival and communication.

PACS1 mutations have been linked to PACS1 Syndrome a neurodevelopmental disorder characterized by delayed development and distinctive facial features. Additionally PACS1 has a role in some cancers likely due to its involvement in cell-cycle progression and apoptosis and it may interact with the p53 tumor suppressor protein in these contexts. Understanding the specific pathways and interactions involving PACS1 could help target therapeutic strategies for these conditions.

Coat protein that is involved in the localization of trans-Golgi network (TGN) membrane proteins that contain acidic cluster sorting motifs. Controls the endosome-to-Golgi trafficking of furin and mannose-6-phosphate receptor by connecting the acidic-cluster-containing cytoplasmic domain of these molecules with the adapter-protein complex-1 (AP-1) of endosomal clathrin-coated membrane pits. Involved in HIV-1 nef-mediated removal of MHC-I from the cell surface to the TGN. Required for normal ER Ca2+ handling in lymphocytes. Together with WDR37, it plays an essential role in lymphocyte development, quiescence and survival. Required for stabilizing peripheral lymphocyte populations (By similarity).
