Palladin is a cytoskeletal protein that plays an important role in maintaining cell structure and function. It is often referred to simply by its name and it has a molecular weight of approximately 90 to 140 kDa depending on its isoform. Palladin is expressed in a variety of tissues but shows a higher presence in cells such as fibroblasts smooth muscle cells and cardiac tissues. This protein interacts with numerous actin-associated proteins and contributes to the organization of the actin cytoskeleton by promoting actin polymerization.
Biological function summary

Palladin interacts with at least four actin-binding proteins to influence cellular processes such as motility and adhesion. Palladin often forms part of larger protein complexes that regulate the cytoskeleton. Through these interactions it contributes to the maintenance of cell shape and the mechanical stability of cells. Palladin’s role extends to influencing cell signaling pathways that regulate cell movement and structural integrity.

Pathways

Researchers connect palladin functions with cytoskeletal organization and cell signaling. It participates in pathways that involve the RhoA signaling pathway and the mechanotransduction pathway. Palladin interacts closely with proteins such as profilin and VASP synergistically influencing actin polymerization. These interactions are important for various cellular activities such as migration and morphogenesis.

Researchers have found palladin to be linked with conditions such as cancer and certain fibrosis. Altered expression levels of palladin can lead to enhanced metastatic potential in cancer cells. Additionally it exhibits associations with BRCA1 in breast cancer where mutations can alter cytoskeletal dynamics thereby affecting cell proliferation and migration. Palladin's overexpression is also observed in pancreatic cancer connecting it to actin-associated proteins that influence disease progression.
