HepG2 (Human liver hepatocellular carcinoma cell line) cells stained for Pannexin 3 (green) using ab237055 at 1/133 dilution in ICC/IF, followed by an Alexa-Fluor®488-conjugated goat anti-rabbit IgG (H+L). Counterstained with DAPI (blue).

The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal goat serum. The cells were then incubated with the antibody overnight at 4°C.

Pannexin 3 also known as Panx3 is a part of the pannexin family of proteins and consists of approximately 43 kDa in mass. Mechanically Panx3 forms large-pore channels that facilitate the passage of ions and small molecules across the cell membrane contributing to intercellular communication. This protein is expressed not only in a range of tissues such as the skin bone and cartilage but also in organs like the heart and liver signaling its widespread influence in the body.
Biological function summary

The protein functions at the cellular level include regulation of calcium signaling and ATP release which influences various cell functions. Panx3 often exists as a part of complexes with other similar proteins like connexins which play roles in forming gap junctions. These activities are essential in controlling cellular interactions and maintaining tissue function particularly in supporting cartilage and bone homeostasis.
