Sandwich ELISA of ab315130 with the capture antibody dilution at 2 µg/mL and detector antibody dilution at 0.5 µg/mL. Interpolated concentrations of native cleaved PARP1 in human wild type HEK293T cell, PARP1 knockout HEK293T cell, wild type HEK293T cell treated with 1µM staurosporine for 4 hours and PARP1 knockout HEK293T cell treated with 1µM staurosporine for 4 hours based on 50 µg/mL extract loads. The concentrations of cleaved PARP1 were measured in duplicate and interpolated from the cleaved PARP1 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean cleaved PARP1 concentration was determined to be 2725.1 pg/mL in wild type HEK293T extract (Human wild-type HEK293T cell line ab255449), 9318.2 pg/mL in wild type HEK293T extract treated with staurosporine, undetectable in both PARP1 knockout HEK293T extract (Human PARP1 knockout HEK293T cell line ab266598) untreated or treated with staurosporine.

Biological function summary
The enzymatic function of PARP1 involves the transfer of ADP-ribose units from NAD+ to target proteins a process known as ADP-ribosylation. PARP1 operates as a part of the base excision repair complex essential in DNA repair processes. The cleaved form of PARP1 no longer facilitates DNA repair marking a shift towards apoptosis. When PARP1 is cleaved it indicates caspase activity implying cells are undergoing programmed cell death.
Pathways
Cleaved PARP1 is deeply involved in the apoptosis and DNA damage response pathways. In the apoptosis pathway PARP1 interacts with key proteins like caspase-3 which cleaves PARP during apoptosis. In the DNA damage response PARP1 collaborates with proteins such as XRCC1 facilitating the base excision repair pathway important for fixing single-strand DNA breaks. These pathways highlight the dual role of PARP1 in promoting cell survival through repair and cell death via apoptosis.
