Supplementary information

PATJ also known as PALS1-associated tight junction protein plays an important role in maintaining cell polarity and tight junction assembly. With a molecular mass of approximately 200 kDa PATJ expresses in various epithelial and endothelial tissues where it contributes to the stability and function of these barriers. This protein localizes at the tight junctions of cells and interacts with other tight junction components to support cellular structure and function.
Biological function summary

PATJ assists in establishing and maintaining cell-cell junction integrity by participating in complexes that involve the Crumbs complex. It supports the correct positioning and functioning of tight junctions important for proper cell adhesion and epithelial cell polarity. Through its interaction with other tight junction proteins like PALS1 and Crumbs PATJ ensures that epithelial cells maintain their differentiated state and modulate paracellular permeability.

Pathways

Cell-cell adhesion and polarity maintenance significantly involve PATJ. It intersects with the Crumbs polarity complex pathway where Crumbs3 PALS1 and PATJ interact to regulate epithelial cell polarity. Additionally PATJ participates with the Par6-aPKC pathway both critical for maintaining apical-basal polarity and tight junctions in epithelial tissues ensuring proper biological function and tissue organization.

PATJ's role in maintaining cell polarity links it to disorders such as cancer and ocular diseases. In certain cancers disrupted expression or function of PATJ may lead to disordered cell polarity and contribute to tumorigenesis. Conditions like retinopathy also implicate PATJ as it functions alongside Crumbs proteins essential for retinal integrity. Understanding PATJ's interactions with these proteins could provide insight into disease mechanisms and potential therapeutic targets.

Target data

Scaffolding protein that facilitates the localization of proteins to the cell membrane (PubMed : 11927608, PubMed : 16678097, PubMed : 22006950). Required for the correct formation of tight junctions and epithelial apico-basal polarity (PubMed : 11927608, PubMed : 16678097). Acts (via its L27 domain) as an apical connector and elongation factor for multistranded TJP1/ZO1 condensates that form a tight junction belt, thereby required for the formation of the tight junction-mediated cell barrier (By similarity). Positively regulates epithelial cell microtubule elongation and cell migration, possibly via facilitating localization of PRKCI/aPKC and PAR3D/PAR3 at the leading edge of migrating cells (By similarity). Plays a role in the correct reorientation of the microtubule-organizing center during epithelial migration (By similarity). May regulate the surface expression and/or function of ASIC3 in sensory neurons (By similarity). May recruit ARHGEF18 to apical cell-cell boundaries (PubMed : 22006950).
