Immunocytochemistry/ Immunofluorescence - Anti-PAX3 antibody [RP23040034] (AB307998)
For immunofluorescence analysis, HeLa cells were stained for detection of PAX3 using ab307998 followed by detection using an Alexa Fluor 488-conjugated Goat anti-Rabbit secondary antibody (panel a : green) Nuclei were stained using DAPI (panel b : blue) Actin stained with Alexa Fluor 594 phalloidin (panel c : red). Panel D is a composite image showing nuclear localization of Pax 3.

Western blot analysis of PAX3 in A431 whole cell extracts using ab307998 at a dilution of 1 µg/mL. Samples were detected using chemiluminescence (ECL). Results show a band at ~53kDa.

Western blot analysis was performed on whole cell extracts of HeLa (Lane 1) and A431 (Lane 2). The blot was probed with ab307998 at 2 µg/mL and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Secondary Antibody, HRP conjugate at 1/2500 dilution. A 53 kDa band corresponding to PAX3 was observed across the cell lines tested. Known quantity of protein samples were electrophoresed using 12 % Bis-Tris gel, electrophoresis system and pre-stained protein standard. Resolved proteins were then transferred onto a nitrocellulose membrane. The membrane was probed with the relevant primary and secondary Antibody. Chemiluminescent detection was performed (ECL).
