PBA1 also known as proteasome subunit beta type-4 or PSMA4 is a component of the 20S proteasome. It is a catalytic core subunit of the proteasome complex and is responsible for the breakdown of proteins through proteolysis. PBA1 has a molecular mass of around 29 kDa and shows a wide expression pattern across tissues. It plays an essential role in the ubiquitin-proteasome system which is central to protein homeostasis within all eukaryotic cells.
Biological function summary

This proteasome subunit is involved in degrading polyubiquitinated proteins thereby regulating various cellular processes. PBA1 is part of the 20S core particle of the proteasome complex which also includes the 19S regulatory particle. This complex regulates protein degradation and turnover impacting cell cycle progression signal transduction and stress responses. PBA1's contribution to protein degradation underlines its importance in maintaining cellular function and preventing the buildup of damaged or misfolded proteins.

Pathways

The role of PBA1 is key in the ubiquitin-proteasome pathway allowing the clearance of unwanted proteins which is fundamental for cellular regulation. This pathway is linked to various biological processes such as cell cycle control and apoptosis. PBA1 interacts with other proteasome subunits like PSMB5 within this pathway to facilitate its proteolytic activity. Additionally PBA1 is associated with the NF-κB signaling pathway where it aids in the activation of transcription factors critical for immune responses and inflammation.

PBA1 has been implicated in neurodegenerative diseases such as Parkinson’s disease due to its role in protein quality control. Impairments in PBA1 function or mutations could contribute to the accumulation of neurotoxic proteins exacerbating disease progression. Moreover abnormalities in proteasome-mediated protein degradation also link PBA1 to various types of cancer including multiple myeloma where it has associations with proteins like ubiquitin that are significant in disease mechanisms.
