Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of PC1/3 using ab191452 in human colon cancer tissues. Heat mediated antigen retrieval was performed in citrate buffer (pH6, epitope retrieval solution ) for 20 mins. The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1μg/mL ab191452 overnight at 4°C. DyLight®488 Conjugated Avidin was used and incubated for 30 minutes at 37°C. The section was counterstained with DAPI. Visualize using a fluorescence microscope and filter sets appropriate for the label used.

PC1/3 also known as Proprotein Convertase 1 or PCSK1 is a member of the proprotein convertase family which is responsible for processing precursor proteins into their active forms. This protein with a mass of approximately 84 kDa is expressed in neuroendocrine and endocrine tissues. PC1/3 operates primarily in the secretory pathways of these tissues cleaving prohormones and other precursor proteins at specific sites to generate active peptides and proteins essential for various physiological processes.
Biological function summary

The proprotein convertase enzyme PC1/3 plays a critical role in activating hormone precursors. It is involved in the conversion of proinsulin to insulin as well as the processing of proopiomelanocortin (POMC) to generate peptides like ACTH and beta-endorphin. PC1/3 does not generally form part of a complex but interacts closely with other convertases for complete prohormone processing. This enzyme is essential for maintaining homeostasis in hormone regulation and is an important player in metabolic control.

Pathways

Specific functions of PC1/3 are closely tied to metabolic and neuroendocrine pathways.
