The PCBP1 protein also known as Poly(rC) Binding Protein 1 or hnRNP E1 is a RNA-binding protein with a mass of approximately 39-40 kDa. Mechanically it binds to single-stranded poly(C) regions in RNA through its three K-homology (KH) domains. These domains allow PCBP1 to regulate mRNA stability translation and splicing. The protein is broadly expressed across many tissues with high levels in the liver thymus kidneys and the brain suggesting its essential involvement in diverse cellular processes.
Biological function summary

PCBP1 influences the cellular machinery through its role in post-transcriptional regulation. It does not function alone but rather exists as part of an RNA-protein complex. This protein participates in modulating mRNA turnover and translational silencing playing important roles in cellular iron homeostasis and response to viral infections. Through its interaction with specific mRNAs PCBP1 helps regulate processes like cell proliferation and differentiation where its dysregulation may have significant consequences.

Pathways

Alterations in PCBP1 activities impact important cellular pathways such as the Wnt signaling pathway and the iron metabolism pathway. In the Wnt signaling pathway PCBP1 interacts with other proteins like beta-catenin to regulate gene expression that guides cell growth and division. In iron metabolism PCBP1 plays a role by stabilizing iron-responsive element (IRE)-containing mRNAs therefore influencing the expression of proteins involved in iron storage and transport.

PCBP1 has connections to cancer and anemia. In various cancers such as colorectal and lung cancer PCBP1 expression levels change suggesting a role in tumor progression and metastasis. It interacts with proteins such as beta-catenin which is known to contribute to oncogenic pathways. In conditions of anemia particularly sideroblastic anemia PCBP1 influences the expression of proteins that are critical for iron regulation like ferritin affecting iron homeostasis in red blood cells. Understanding these relationships provides potential avenues for therapeutic interventions targeting PCBP1 and associated pathways.
