This data was developed using ab202560, the same antibody clone in a different buffer formulation. Immunofluorescent analysis of 4% paraformaldehyde-fixed, 0.1% Triton X-100 permeabilized HEK-293T (Human epithelial cell line from embryonic kidney transformed with large T antigen) cells transfected with a GFP-tagged mouse PCDH15 expression construct or GFP vector only, labeling PCDH15 with ab202560 at 1/500 dilution, followed by Goat anti-rabbit IgG (Alexa Fluor® 594) (ab150080) secondary antibody at 1/1000 dilution (red).

This mouse PCDH15 plasmid was kindly provided by our collaborator Dr. Yun Shi, Nanjing University MARC.

The nuclear counterstain is DAPI (blue).

Tubulin is detected with ab7291 (anti-Tubulin mouse mAb) at 1/1000 dilution and ab150115 (Alexa Fluor ® 647 Goat anti-Mouse secondary) at 1/1000 dilution (white).

Secondary antibody only control : Used PBS instead of primary antibody, secondary antibody is Goat anti-rabbit IgG (Alexa Fluor® 594) (ab150080) at 1/1000 dilution.

PCDH15 also known as Protocadherin-15 is a protein encoded by the PCDH15 gene. Mechanically this protein functions as an adhesion molecule in the body playing an important role in cell-cell interactions. PCDH15 has a molecular mass of approximately 223 kDa. It has a widespread expression but is particularly found in sensory cells in the inner ear and in the retina of the eye highlighting its importance in sensory functions.
Biological function summary

PCDH15 participates in the formation of tip-link structures within the inner ear. This protein is a component of the mechanotransduction complex which translates mechanical stimuli into electrical signals. PCDH15 works alongside other proteins like Cadherin 23 another protocadherin to maintain the integrity of hair cells. Its role is essential for proper auditory and visual signal processing contributing to the sense of hearing and vision.

Pathways

PCDH15 interacts with the mechanotransduction pathway in sensory cells. This pathway plays a role in converting mechanical stimuli such as sound waves in the cochlea into nerve impulses. Cadherin 23 is closely related in this pathway and together these proteins maintain the hair cell structure necessary for mechanotransduction. The interplay between these proteins ensures the transduction of sensory information impacting cellular responses in sensory organs.

PCDH15 mutations associate with Usher syndrome type 1F and non-syndromic hearing loss. Usher syndrome is a genetic condition involving hearing loss and vision impairment. PCDH15 mutations disrupt the mechanotransduction complex affecting sensory perception. Cadherin 23 also connects to these disorders as mutations in this protein can similarly lead to issues in hearing and vision. Understanding PCDH15 and its interactors is critical in addressing these sensory disorders.
