The protein PCDH18 also known as Protocadherin 18 is part of the protocadherin family which consists of diverse cell-adhesion molecules. PCDH18 has a known mass of approximately 115 kDa. It plays a significant role in cell-cell communication and is mainly expressed in the brain particularly in the developing neural tissues. Its expression in neural tissues suggests it participates in the intricate processes involved in the development and maintenance of neuronal connections.
PCDH18 acts as a cell adhesion molecule contributing to the establishment and maintenance of specific cell-cell contacts. It does not appear to be part of a larger complex but interacts with other cell adhesion molecules to influence neuronal networking and synaptic integrity. This protein aids in the regulation of neural circuit assembly and plasticity which are essential for proper brain function and cognitive processes.
The role of PCDH18 in neural connections fits within key neuronal signaling pathways. One major pathway is the Wnt signaling pathway which influences cell movement and polarity in the developing nervous system. PCDH18 interacts with other proteins involved in this pathway such as beta-catenin to modulate signaling that affects neuronal patterning and synapse formation. Furthermore its interaction within cadherin-related pathways connects it with proteins like N-cadherin which collaborates in neural development and synapse configuration.
The function of PCDH18 links to two main neurological conditions: schizophrenia and autism spectrum disorders. Alterations in PCDH18 expression can disrupt synaptic functions leading to these cognitive and social interaction impairments. Additionally PCDH18 relates with other proteins involved in these disorders such as Neurexin and Neuroligin which are important in synapse maintenance and neural circuitry. Understanding these connections offers potential pathways for therapeutic targets in managing these neurological conditions.
