Supplementary information

PCGF3 also known as Polycomb Group RING Finger 3 is a protein that plays a mechanical role as part of the Polycomb group (PcG) proteins. The molecular mass of PCGF3 typically ranges around 35 kDa. It is expressed in various tissues with a notable presence in embryonic stem cells and during early developmental stages. PCGF3 functions as a component of Polycomb Repressive Complex 1 (PRC1) and influences chromatin modification impacting gene expression.
Biological function summary

PCGF3 influences gene silencing and epigenetic regulation. This protein forms part of the PRC1 complex which is important for maintaining the transcriptionally repressive state of many genes throughout development. PCGF3 alongside other PcG proteins helps to regulate chromatin structure by mediating the mono-ubiquitination of histone H2A one of its key functions in chromatin modification.

Pathways

The PCGF3 protein takes part in the Polycomb pathway which impacts the transcriptional silencing of Hox genes critical for body patterning and development. It relates closely to other proteins within this pathway such as BMI1 which is also a member of the PRC1 complex. Additionally PCGF3 plays a role in the regulation of the WNT signaling pathway influencing cellular differentiation and proliferation.

PCGF3 has links to various cancers and developmental disorders given its role in gene silencing and epigenetic regulation. Changes in its expression can lead to aberrant activation or repression of target genes potentially contributing to tumorigenesis. Research indicates that alterations in WNT signaling involving PCGF3 and related proteins such as RING1B could play a role in the pathogenesis of colorectal cancer and other malignancies.

Target data

Component of a Polycomb group (PcG) multiprotein PRC1-like complex, a complex class required to maintain the transcriptionally repressive state of many genes, including Hox genes, throughout development. PcG PRC1 complex acts via chromatin remodeling and modification of histones; it mediates monoubiquitination of histone H2A 'Lys-119', rendering chromatin heritably changed in its expressibility. Within the PRC1-like complex, regulates RNF2 ubiquitin ligase activity (PubMed : 26151332). Plays a redundant role with PCGF5 as part of a PRC1-like complex that mediates monoubiquitination of histone H2A 'Lys-119' on the X chromosome and is required for normal silencing of one copy of the X chromosome in XX females (By similarity).
