Immunofluorescent analysis of 4% paraformaldehyde-fixed, 0.1% Triton X-100 permeabilized F9 (Mouse embryonic testicular cancer cell line) cells labeling PCGF6 with ab200038 at 1/100 dilution, followed by Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) (ab150077) secondary antibody at 1/1000 dilution (green). Tubulin is detected with Anti-alpha Tubulin mouse MAb (ab7291) at 1/1000 dilution, followed by Goat Anti-Mouse IgG H&L (Alexa Fluor® 594) (ab150120) secondary antibody at 1/1000 dilution (red).

ab200038 Immunoprecipitating PCGF6 in F9 (mouse embryonal carcinoma) whole cell lysate. 350μg of cell lysate was incubated with primary antibody (2 μg/ml). For western blotting, ab200038 (1/1000) was used to confirm successful immunoprecipation. For detection VeriBlot for IP Detection Reagent (HRP) (ab131366) was used at 1/1000.

PCGF6 also known as Polycomb group RING finger protein 6 plays a role in regulating gene expression. This protein with a molecular mass of about 36 kDa is part of the Polycomb Group (PcG) proteins that epigenetically silence genes preventing their expression. Scientists have observed PCGF6 expression in embryonic stem cells as well as in various tissues hinting at its broad potential impact on cellular processes. The protein contains a RING finger motif which suggests it functions as an E3 ubiquitin ligase tagging other proteins for modification or degradation.

Component of a Polycomb group (PcG) multiprotein PRC1-like complex, a complex class required to maintain the transcriptionally repressive state of many genes, including Hox genes, throughout development. PcG PRC1 complex acts via chromatin remodeling and modification of histones; it mediates monoubiquitination of histone H2A 'Lys-119', rendering chromatin heritably changed in its expressibility.
