PDE3A also known as phosphodiesterase 3A regulates the levels of cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP) in cells by hydrolyzing these cyclic nucleotides. This enzyme with a molecular mass of approximately 133 kDa is expressed in tissues including the heart vascular smooth muscle and platelets. PDE3A activity influences contractility and promotes cell cycling and differentiation through its role in modulating signal transduction pathways.
Biological function summary

Phosphodiesterase 3A plays an important role in cardiac and vascular functions. It forms part of a signaling complex responsible for regulating cardiovascular responses. PDE3A manages intracellular cAMP and cGMP concentrations impacting vascular smooth muscle relaxation and platelet aggregation. The modulation of these signaling molecules by PDE3A contributes to maintaining normal heart rhythms and influencing blood pressure.

Pathways

Phosphodiesterase 3A is central to the cAMP and cGMP signaling pathways. PDE3A activity connects to the cardiac contractility pathway where it works alongside proteins like protein kinase A (PKA) which depends on cAMP levels. Research indicates that PDE3A contributes to cardiovascular conditions like heart failure and hypertension. In heart failure altered PDE3A activity can upset cAMP levels impacting cardiac contractility. Other proteins such as PKA in cardiac tissue and PKG in vascular tissues are involved in these conditions and their regulation by PDE3A adds another layer to understanding these diseases.

Cyclic nucleotide phosphodiesterase with specificity for the second messengers cAMP and cGMP, which are key regulators of many important physiological processes (PubMed : 1315035, PubMed : 25961942, PubMed : 8155697, PubMed : 8695850). Has also activity toward cUMP (PubMed : 27975297). Independently of its catalytic activity it is part of an E2/17beta-estradiol-induced pro-apoptotic signaling pathway. E2 stabilizes the PDE3A/SLFN12 complex in the cytosol, promoting the dephosphorylation of SLFN12 and activating its pro-apoptotic ribosomal RNA/rRNA ribonuclease activity. This apoptotic pathway might be relevant in tissues with high concentration of E2 and be for instance involved in placenta remodeling (PubMed : 31420216, PubMed : 34707099).
