Detection of PDE3A in Immunoprecipitates of HeLa whole cell lysates (1 mg for IP, 20% of IP loaded) using ab99236 at 3 µg/mg lysate for IP (Lane 1) and at 1 µg/ml for subsequent Western blot detection. Lane 2 represents control IgG IP. Detection : Chemiluminescence with an exposure time of 3 seconds.

In Western blot, ab99236 was shown to bind specifically to PDE3A. First, samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. Membranes were blocked in 3 % milk in TBS-0.1 % Tween® 20 (TBS-T) before incubation with primary antibodies overnight at 4 °C. Blots were washed four times in TBS-T, incubated with secondary antibodies for 1 h at room temperature, washed again four times then imaged. Secondary antibodies used were Goat anti-Rabbit IgG H&L (IRDye® 800CW) preabsorbed (ab216773) and Goat anti-Mouse IgG H&L (IRDye® 680RD) preabsorbed (ab216776) at 1/20000 dilution.

PDE3A also known as phosphodiesterase 3A regulates the levels of cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP) in cells by hydrolyzing these cyclic nucleotides. This enzyme with a molecular mass of approximately 133 kDa is expressed in tissues including the heart vascular smooth muscle and platelets. PDE3A activity influences contractility and promotes cell cycling and differentiation through its role in modulating signal transduction pathways.
Biological function summary

Cyclic nucleotide phosphodiesterase with specificity for the second messengers cAMP and cGMP, which are key regulators of many important physiological processes (PubMed : 1315035, PubMed : 25961942, PubMed : 8155697, PubMed : 8695850). Also has activity toward cUMP (PubMed : 27975297). Independently of its catalytic activity it is part of an E2/17beta-estradiol-induced pro-apoptotic signaling pathway. E2 stabilizes the PDE3A/SLFN12 complex in the cytosol, promoting the dephosphorylation of SLFN12 and activating its pro-apoptotic ribosomal RNA/rRNA ribonuclease activity. This apoptotic pathway might be relevant in tissues with high concentration of E2 and be for instance involved in placenta remodeling (PubMed : 31420216, PubMed : 34707099).
