Western blot - Anti-PDK1 antibody [EPR13015] - C-terminal (AB180171)

Lane 1 : Wild-type HAP1 cell lysate (20 μg)
Lane 2 : Mitochondrial Pyruvate dehydrogenase kinase 1 knockout HAP1 cell lysate (20 μg)
Lane 3 : Human fetal heart tissue lysate (20 μg)
Lane 4 : LnCaP cell lysate (20 μg)
Lanes 1 - 4 : Merged signal (red and green). Green - ab180171 observed at 46 kDa. Red - loading control, ab8245, observed at 37 kDa.

ab180171 was shown to specifically react with Mitochondrial Pyruvate dehydrogenase kinase 1 in wild-type HAP1 cells along with additional cross reactive bands. No bands were observed when Mitochondrial Pyruvate dehydrogenase kinase 1 knockout samples were examined. Wild-type and Mitochondrial Pyruvate dehydrogenase kinase 1 knockout samples were subjected to SDS-PAGE. ab180171 and ab8245 (loading control to GAPDH) were both diluted 1/1000 and incubated overnight at 4°C. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preadsorbed ab216773 and Goat anti-Mouse IgG H&L (IRDye® 680RD) preadsorbed ab216776 secondary antibodies at 1/10,000 dilution for 1 hour at room temperature before imaging.

Target data

Kinase that plays a key role in regulation of glucose and fatty acid metabolism and homeostasis via phosphorylation of the pyruvate dehydrogenase subunits PDHA1 and PDHA2 (PubMed : 7499431, PubMed : 18541534, PubMed : 22195962, PubMed : 26942675, PubMed : 17683942). This inhibits pyruvate dehydrogenase activity, and thereby regulates metabolite flux through the tricarboxylic acid cycle, down-regulates aerobic respiration and inhibits the formation of acetyl-coenzyme A from pyruvate (PubMed : 18541534, PubMed : 22195962, PubMed : 26942675). Plays an important role in cellular responses to hypoxia and is important for cell proliferation under hypoxia (PubMed : 18541534, PubMed : 22195962, PubMed : 26942675).
