Function

Kinase that plays a key role in regulation of glucose and fatty acid metabolism and homeostasis via phosphorylation of the pyruvate dehydrogenase subunits PDHA1 and PDHA2. This inhibits pyruvate dehydrogenase activity, and thereby regulates metabolite flux through the tricarboxylic acid cycle, down-regulates aerobic respiration and inhibits the formation of acetyl-coenzyme A from pyruvate. Plays an important role in cellular responses to hypoxia and is important for cell proliferation under hypoxia. Protects cells against apoptosis in response to hypoxia and oxidative stress.

    Lane 2: Mitochondrial Pyruvate dehydrogenase kinase 1 knockout HAP1 cell lysate (20 μg)

    Lanes 1 - 4: Merged signal (red and green). Green - ab180171 observed at 46 kDa. Red - loading control, Anti-GAPDH antibody [6C5] - Loading Control ab8245, observed at 37 kDa.

    ab180171 was shown to specifically react with Mitochondrial Pyruvate dehydrogenase kinase 1 in wild-type HAP1 cells along with additional cross reactive bands. No bands were observed when Mitochondrial Pyruvate dehydrogenase kinase 1 knockout samples were examined. Wild-type and Mitochondrial Pyruvate dehydrogenase kinase 1 knockout samples were subjected to SDS-PAGE. ab180171 and Anti-GAPDH antibody [6C5] - Loading Control ab8245 (loading control to GAPDH) were both diluted 1/1000 and incubated overnight at 4°C. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preadsorbed Goat anti-Rabbit IgG H&L (IRDye® 800CW) preadsorbed ab216773 and Goat anti-Mouse IgG H&L (IRDye® 680RD) preadsorbed Goat anti-Mouse IgG H&L (IRDye® 680RD) preadsorbed ab216776 secondary antibodies at 1/10,000 dilution for 1 hour at room temperature before imaging.
