Kinase that plays a key role in regulation of glucose and fatty acid metabolism and homeostasis via phosphorylation of the pyruvate dehydrogenase subunits PDHA1 and PDHA2. This inhibits pyruvate dehydrogenase activity, and thereby regulates metabolite flux through the tricarboxylic acid cycle, down-regulates aerobic respiration and inhibits the formation of acetyl-coenzyme A from pyruvate. Plays an important role in cellular responses to hypoxia and is important for cell proliferation under hypoxia. Protects cells against apoptosis in response to hypoxia and oxidative stress.

The PDK1 protein also known as 3-phosphoinositide-dependent protein kinase-1 is a kinase enzyme coded by the PDPK1 gene. PDK1 has a molecular weight of approximately 63 kDa. It is expressed across a variety of tissues including high levels in the heart pancreas liver skeletal muscle and in certain brain regions. This kinase plays a pivotal role in cellular signaling pathways due to its ability to phosphorylate other proteins.
PDK1 acts as a master regulator of the AGC family of kinases which includes proteins like PKB/Akt S6K and SGK. By phosphorylating these proteins PDK1 controls cell growth glycogen synthesis and apoptosis. It does not function within a complex but operates as an important activator in phosphorylation events influencing several downstream cellular responses. By modulating kinase activity PDK1 significantly impacts cellular metabolism and survival pathways.
PDK1 serves as a critical component in the PI3K/Akt signaling pathway and the mTOR pathway. In the PI3K/Akt pathway PDK1 phosphorylates and activates Akt leading to varied outcomes such as glucose uptake cell proliferation and resistance to apoptosis under stressful conditions. The relationship between PDK1 and other proteins like mTOR highlights its importance in regulating cell growth and survival through nutrient-sensing mechanisms.
PDK1 is heavily implicated in cancer development and diabetes. Aberrant activation of the PI3K/Akt/mTOR pathway often driven by mutations influencing PDK1 activity is a common feature in many cancers. PDK1's role in modulating insulin signaling also connects it to type 2 diabetes where insulin resistance is a characteristic issue. Through these pathways PDK1 interacts with oncogenes and proteins like PTEN further influencing disease progression and treatment outcomes.

Anti-PDK1 antibody [EPR19573] ab207450 was shown to react with PDK1 in wild-type HeLa cells in western blot. Loss of signal was observed when knockout cell line ab264791 (knockout cell lysate ab257164) was used. Wild-type HeLa and PDK1 knockout HeLa cell lysates were subjected to SDS-PAGE. Membrane was blocked for 1 hour at room temperature in 0.1% TBST with 3% non-fat dried milk. Anti-PDK1 antibody [EPR19573] ab207450 and Anti-GAPDH antibody [6C5] - Loading Control (Anti-GAPDH antibody [6C5] - Loading Control ab8245) overnight at 4°C at a 1 in 2000 dilution and a 1 in 20000 dilution respectively. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye®800CW) preadsorbed (Goat anti-Rabbit IgG H&L (IRDye® 800CW) preadsorbed ab216773) and Goat anti-Mouse IgG H&L (IRDye®680RD) preadsorbed (Goat anti-Mouse IgG H&L (IRDye® 680RD) preadsorbed ab216776) secondary antibodies at 1 in 20000 dilution for 1 hour at room temperature before imaging.
