Supplementary information

PDLIM5/ENH also known as Enhancer of Lim Domain Protein 5 is a cytoskeletal protein with a molecular mass of approximately 62 kDa. This protein is primarily expressed in heart and skeletal muscle tissues. PDLIM5/ENH binds to actin filaments and is involved in organizing the cytoskeleton. It acts as an adapter or scaffold protein linking other proteins to the cytoskeleton and helping in maintaining structural integrity and function in muscle cells.
Biological function summary

PDLIM5/ENH plays a significant role in cardiac muscle development and function. It is also a part of the protein complex that interacts with other cytoskeletal proteins to stabilize actin filament structures. This interaction is important for muscle contraction and cellular signaling. PDLIM5/ENH is also involved in transducing mechanical signals into biochemical signals which affect cell differentiation and growth.

Pathways

PDLIM5/ENH integrates into important signaling pathways like the MAPK and Wnt pathways. In the MAPK pathway PDLIM5/ENH interacts with proteins involving cell growth and differentiation such as ERK. Through the Wnt pathway it plays a role in tissue development and regeneration by working alongside proteins like beta-catenin. These pathways highlight its contribution to essential cellular processes that affect muscle and cardiac function.

PDLIM5/ENH is associated with cardiac hypertrophy and dilated cardiomyopathy. Alterations in its function can impact cardiac muscle structure leading to pathological changes. Additionally its interaction with proteins like desmin in muscle tissues links it to muscle disorder pathologies. Understanding PDLIM5/ENH's role in these diseases may provide insights into therapeutic targets for cardiac and muscle-related disorders.

Target data

May play an important role in the heart development by scaffolding PKC to the Z-disk region. May play a role in the regulation of cardiomyocyte expansion. Isoforms lacking the LIM domains may negatively modulate the scaffolding activity of isoform 1. Overexpression promotes the development of heart hypertrophy. Contributes to the regulation of dendritic spine morphogenesis in neurons. May be required to restrain postsynaptic growth of excitatory synapses. Isoform 1, but not isoform 2, expression favors spine thinning and elongation.
