PDZ Binding Kinase (PBK) also known as TOPK is a serine/threonine kinase with a molecular mass of approximately 36 kDa. It performs phosphorylation on certain proteins influencing their function localization or interaction with other proteins. PBK locates mainly in the testes placenta and various tumors but also expresses in smaller amounts in normal tissues such as the liver and brain. The kinase is cell cycle-dependent with activity peaking during mitosis where it plays a role by interacting with the mitotic spindle apparatus.

PBK acts as part of cellular proliferation and differentiation processes. It can form complexes with other proteins enhancing its function and stability. Phosphorylates MAP kinase p38. Seems to be active only in mitosis. May also play a role in the activation of lymphoid cells. When phosphorylated, forms a complex with TP53, leading to TP53 destabilization and attenuation of G2/M checkpoint during doxorubicin-induced DNA damage.
