Flow Cytometry - PE Anti-CD130 (gp130) antibody [EPR24557-5] (AB322916)
Flow cytometry overlay histogram showing wild-type A549 (green line) and CD130 knockout A549 stained with ab322916 (magenta line). The cells were fixed with 80% methanol (5 min) and then permeabilised with 0.1% PBS-Triton X-100 for 15 min. The cells were incubated in 1x PBS containing 10% normal goat serum to block non-specific protein-protein interaction followed by the antibody (ab322916) (1x 106 in 100 μl at 0.2 μg/ml (1/2500)) for 30min at 22°C.
Isotype control antibody Rabbit IgG (monoclonal) Phycoerythrin Isotype Control (ab209478) was used at the same concentration and conditions as the primary antibody (wild-type A549 - black line, CD130 knockout A549 - grey line). Unlabelled sample was also used as a control (this line is not shown for the purpose of simplicity).
Acquisition of >5000 events were collected using a 50 mW Yellow/Green laser (561nm) and 585/42 bandpass filter.
This antibody gave a positive signal in cells fixed with 4% formaldehyde (10 min) / permeabilised with 0.1% PBS-Triton X-100 for 15 min under the same conditions.

Biological function summary
CD130 functions as part of the high-affinity receptor complex for certain cytokines. It partners with other receptors such as CD126 to form a complex that initiates signaling pathways after ligand binding contributing to a range of immune responses. Its role is thorough across many cellular processes including differentiation proliferation and apoptosis. In addition to immune signaling CD130 is involved in several growth factor signaling pathways.
Pathways
CD130 serves a significant role in signal transduction particularly in the Janus kinase/signal transducers and activators of transcription (JAK/STAT) pathway. It works in concert with related proteins like JAK1 and STAT3 to transmit signals from extracellular cytokines to the nucleus influencing gene expression. Additionally CD130 can also be found active in the MAPK/ERK pathway which is important for cellular response to growth signals.
