Flow Cytometry (Intracellular) - PE Anti-GPCR LGR6 antibody [EPR6874] (AB314286)

Flow cytometry overlay histogram showing HeLa cells stained with ab314286 (red line). The cells were fixed with 4% formaldehyde (10 min) and then permeabilised with 0.1% PBS-Triton X-100 for 15 min. The cells were incubated in 1x PBS containing 10% normal goat serum to block non-specific protein-protein interaction followed by the antibody (ab314286) (1x 106 in 100μl at 0.04μg/ml (1/12500)) for 30min at 22°C.

Isotype control antibody (black line) was Rabbit IgG (monoclonal) Phycoerythrin Isotype Control (ab209478) used at the same concentration and conditions as the primary antibody. Unlabelled sample (blue line) was also used as a control.

Acquisition of >5000 events were collected using a 50 mW Yellow/Green laser (561nm) and 585/42 bandpass filter.

This antibody gave a positive signal in HeLa fixed with 80% methanol (5 min) / permeabilised with 0.1% PBS-Triton X-100 for 15 min under the same conditions.

GPCR LGR6 also known as leucine-rich repeat-containing G protein-coupled receptor 6 is a transmembrane receptor that belongs to the large GPCR family. It has a molecular weight of approximately 98 kDa. LGR6 expresses in various tissues including skin bone and certain regions of the brain. The receptor binds specific ligands which initiates signal transduction pathways that mediate various cellular responses. This function places LGR6 as an important component in multiple physiological processes.
Biological function summary

LGR6 plays an important role in signaling pathways involved in stem cell maintenance and tissue regeneration. It functions together with other proteins as part of a larger receptor complex. In the skin LGR6 marks a population of stem cells involved in hair follicle cycling and epidermal renewal. Its activity influences the cellular communication required for tissue homeostasis and repair. This receptor helps in determining cellular fate and proliferation in various tissues.

Pathways

LGR6 participates in the Wnt signaling pathway which is important for cell differentiation growth and migration. LGR6 interacts with proteins such as R-spondins enhancing Wnt signaling. This connection highlights LGR6's role in modulating pathways essential for embryonic development and tissue patterning. The interaction with Wnt and related proteins positions LGR6 as a modulator of pathways that govern stem cell behavior.

Abnormalities in LGR6 function have associations with skin disorders and cancers. Mutations or dysregulation of LGR6 can lead to impaired wound healing and contribute to pathological conditions. Target data

Receptor for R-spondins that potentiates the canonical Wnt signaling pathway and acts as a marker of multipotent stem cells in the epidermis. Upon binding to R-spondins (RSPO1, RSPO2, RSPO3 or RSPO4), associates with phosphorylated LRP6 and frizzled receptors that are activated by extracellular Wnt receptors, triggering the canonical Wnt signaling pathway to increase expression of target genes. In contrast to classical G-protein coupled receptors, does not activate heterotrimeric G-proteins to transduce the signal. May act as a tumor suppressor.
