Pellino 1 and Pellino 2 are E3 ubiquitin ligases and known as PELI1 and PELI2 respectively. PELI1 has a mass of approximately 42 kDa while PELI2 has a similar size around 40 kDa. These proteins are expressed in various tissues including immune cells where they play important roles in cellular signaling processes. Both Pellino proteins contain RING-like domains which facilitate their role in protein ubiquitination a process tagging proteins for degradation or modulating their function.
Biological function summary

Pellino proteins are involved in immune response modulation. They influence signaling pathways such as Toll-like receptor (TLR) and interleukin-1 receptor (IL-1R) pathways. Pellino proteins participate in forming complexes with other signaling proteins assisting in the propagation of inflammatory responses. Their ability to link with components like IRAK1 and TRAF6 helps regulate these immune signaling cascades critical for innate immunity.

Pathways

Pellino proteins are involved in the TLR and IL-1R pathways which are key to innate immune responses. These pathways often include the activation of NF-kB a major transcription factor that regulates immune and inflammatory responses. Pellino proteins interact with signaling proteins like IRAK1 leading to the further propagation of signals that culminate in the activation of NF-kB and the production of pro-inflammatory cytokines.

Pellino proteins are connected to conditions like rheumatoid arthritis and systemic lupus erythematosus. In rheumatoid arthritis aberrant signaling pathways involving Pellino proteins contribute to chronic inflammation and tissue damage. Through their interaction with IRAK1 Pellino proteins can affect downstream signaling that exacerbates auto-inflammatory responses. Their dysregulation can therefore link directly to mechanisms underpinning these autoimmune diseases.
