Pellino 2 also known as PELI2 is an E3 ubiquitin ligase. It is about 45 kDa in mass and exists in the cytoplasm. This protein plays an important role in the regulation of the innate immune response. It is highly expressed in immune cells like monocytes and macrophages as well as in tissues such as the spleen and lymph nodes. As an adaptor molecule Pellino 2 modifies proteins by attaching ubiquitin which tags them for degradation or alters their activity.
Biological function summary

Pellino 2 engages in downstream signaling of immune receptors. It has a critical role in the activation of transcription factors contributing to the production of pro-inflammatory cytokines. Acting in a complex with other components like TRAF proteins it regulates the nuclear factor kappa B (NF-kB) signaling pathway. Through this regulation Pellino 2 influences inflammatory responses and aids in controlling the duration and intensity of inflammation.

Pellino 2 integrates into the Toll-like receptor (TLR) and interleukin-1 (IL-1) signaling pathways. These pathways are fundamental for initiating the innate immune response upon detecting pathogens. Involved in the TLR and IL-1 signaling pathways via interaction with the complex containing IRAK kinases and TRAF6. Mediates IL1B-induced IRAK1 'Lys-63'-linked polyubiquitination and possibly 'Lys-48'-linked ubiquitination. May be important for LPS- and IL1B-induced MAP3K7-dependent, but not MAP3K3-dependent, NF-kappa-B activation. Can activate the MAP (mitogen activated protein) kinase pathway leading to activation of ELK1.
