PEPT2 also known as SLC15A2 is a peptide transporter protein critical for the reabsorption of di- and tripeptides. Mechanically it facilitates peptide transport across cell membranes using a proton gradient-driven mechanism. The protein has a molecular mass of approximately 86.5 kDa. PEPT2 is expressed mainly in the kidney and lung tissues where it plays an important role in nutrient absorption and drug pharmacokinetics.
Biological function summary

PEPT2 participates in the reabsorption and retention of small peptides and peptide-like drugs within cells. It acts as a transmembrane protein and is not known to be part of any larger complex. PEPT2 helps maintain peptide homeostasis by mediating the cellular uptake of short peptides derived from dietary proteins which ensures efficient nutrient recycling and impacts peptide metabolism.

Pathways

PEPT2 significantly contributes to cellular uptake in the nitrogen metabolism pathway and influences the pharmacological profiles of peptide-mimetic drugs in the body. It interacts with other peptide transporters like PEPT1 though PEPT2 operates with varying affinity and capacity particularly affecting the renal reabsorption pathways. Both PEPT2 and PEPT1 play a role in maintaining the body's peptide balance and regulating nitrogen compounds.

Researchers have found associations between PEPT2 function and certain disorders like cystinuria and oligopeptide transport defects. The abnormal activity or expression of PEPT2 can alter the absorption and excretion of key peptides leading to imbalance. In cystinuria for example PEPT2 might influence cystine reabsorption connected as well to cystine transport proteins suggesting altered handling of critical nitrogenous compounds in the disorder.

Transports neutral and anionic dipeptides with a proton to peptide stoichiometry of 2 : 1 or 3 : 1 (By similarity).
