Immunofluorescent analysis of 4% Paraformaldehyde-fixed, 0.1% TritonX-100 permeabilized Human PBMC (human peripheral blood mononuclear cell) cells labelling Perforin with ab324491 at 1/100 (10.21 ug/ml) dilution, followed by ab150117 Goat Anti-Mouse IgG H&L (Alexa Fluor® 488) preadsorbed antibody at 1/1000 (2 ug/ml) dilution (Green).

ab313779 Anti-NCAM1(CD56) rabbit monoclonal antibody was used to counterstain tubulin at 1/200 (2.5 ug/ml) dilution, followed by ab150088 Goat Anti-Rabbit IgG H&L (Alexa Fluor® 594) preadsorbed at 1/1000 (2 ug/ml) (Magenta).

Flow cytometric analysis of 2% paraformaldehyde fixed 0.1% Tween-20 permeabilized Human PBMC (human peripheral blood mononuclear cell) cells labelling Perforin with ab324491 at 1/5000 dilution (0.01ug) / Right compared with a Mouse monoclonal IgG / Left isotype control.

Goat anti-Mouse IgG (Alexa Fluor® 488, ab150113) at 1/5000 dilution was used as the secondary antibody.

Cells were stained with anti-CD56 conjugated to Alexa Fluor®647. Then fixed with 2% PFA for 10 min followed by intracellularly staining with mouse IgG or our antibody.

Target data

Pore-forming protein that plays a key role in granzyme-mediated programmed cell death, and in defense against virus-infected or neoplastic cells (PubMed : 20889983, PubMed : 21037563, PubMed : 24558045, PubMed : 9058810, PubMed : 9164947). Plays an important role in killing other cells that are recognized as non-self by the immune system, e.g. in transplant rejection or some forms of autoimmune disease (PubMed : 9058810). Can insert into the membrane of target cells in its calcium-bound form, oligomerize and form large pores (PubMed : 20889983, PubMed : 21037563). Promotes cytolysis and apoptosis of target cells by mediating the passage and uptake of cytotoxic granzymes (PubMed : 20038786, PubMed : 20225066, PubMed : 24558045, PubMed : 32299851). Facilitates the delivery of cationic cargo protein, while anionic or neural proteins are not delivered efficiently (PubMed : 24558045). Perforin pores allow the release of mature caspase-7 (CASP7) into the extracellular milieu (By similarity).
