Essential component of the nuclear pore complex (NPC) that seems to be required for NPC assembly and maintenance (PubMed:12718872). As part of the NPC Nup107-160 subcomplex plays a role in RNA export and in tethering NUP96/Nup98 and NUP153 to the nucleus (PubMed:12718872). The Nup107-160 complex seems to be required for spindle assembly during mitosis (PubMed:16807356). NUP85 is required for membrane clustering of CCL2-activated CCR2 (PubMed:15995708). Seems to be involved in CCR2-mediated chemotaxis of monocytes and may link activated CCR2 to the phosphatidyl-inositol 3-kinase-Rac-lammellipodium protrusion cascade (PubMed:15995708). Involved in nephrogenesis (PubMed:30179222).

Pericentrin also known as FROUNT is a protein that plays a critical role in organizing microtubule arrays inside cells. It exhibits a molecular mass of approximately 360-400 kDa. This protein localizes mainly at the centrosome an important cellular structure involved in microtubule nucleation as well as at basal bodies within cilia. In centrosomes Pericentrin serves as an anchoring point for several other proteins which helps in maintaining centrosome integrity and supporting cell division processes.
Pericentrin helps coordinate the microtubule network facilitating essential cellular events such as mitosis and intracellular transport. As part of a larger protein complex Pericentrin interacts with gamma-tubulin and several other centrosomal proteins to enhance microtubule nucleation. Additionally Pericentrin helps stabilize the microtubule organizing center ensuring proper cell cycle progression and cellular architecture.
Pericentrin plays a role in the centrosome cycle and mitotic spindle assembly. It interacts with proteins like CEP152 and CDK5RAP2 which are important for proper centrosome duplication and function. These interactions contribute to pathways involved in cell division particularly the regulation of the mitotic spindle and chromosome stability during cell division.
Pericentrin mutations or dysfunctions have been linked to conditions such as microcephalic osteodysplastic primordial dwarfism and certain cancers. Its role in centrosome integrity ties it to BRCA1 a protein associated with breast and ovarian cancer susceptibility. Disrupted Pericentrin function can lead to abnormal cell division and aneuploidy which are common features in cancerous growths.
