Biological function summary

Periplakin participates in the structural organization of epithelial cells. It associates with other proteins to form cytoskeletal linker complexes that enhance cellular stability. As a component of the cornified envelope Periplakin interacts with envoplakin and involucrin. These interactions modulate the cross-linking within the cell envelope impacting the barrier function of the skin and mucous membranes.

Activity summary

Periplakin also known as PPL is a protein that plays an important role in cell adhesion and maintaining cellular structures. It is part of the plakin protein family distinguished by their ability to link cytoskeletal elements in cells. The molecular weight of Periplakin is approximately 195 kDa. Expression of Periplakin occurs mainly in epithelial tissues where it contributes to the formation of desmosomes and other adhesion complexes important for tissue integrity.

Pathways

Periplakin involvement influences epithelial tissue differentiation and stability. It acts within the desmosome assembly pathway impacting cell-cell adhesion processes critical for tissue architecture. Within this pathway Periplakin associates with desmoplakin and envoplakin enhancing cell cohesion and resistance to mechanical stress.

Associated diseases and disorders

Periplakin has been associated with skin conditions such as pemphigus and certain types of cancers. In pemphigus an autoimmune disease antibodies target desmosome components leading to weakened cell adhesion. These events can disrupt Periplakin interactions affecting cellular stability. In cancer altered expression of Periplakin might impact cellular cohesion and migration. Such alterations in association with proteins like envoplakin and involucrin may contribute to disease progression.
