Pescadillo also known as Pes1 is a nucleolar protein with an approximate molecular mass of 110 kDa. It is expressed in the nucleolus of human cells and plays a significant mechanical role in ribosome biogenesis. The protein functions in the processing of the 47S pre-rRNA transcript into 18S and 28S rRNAs which are essential components of the small and large ribosomal subunits respectively. Pescadillo is also involved in cell proliferation indicated by its higher expression levels in rapidly dividing cells such as cancer cells.
Biological function summary

The function of Pescadillo extends beyond ribosome production. It forms a part of a larger complex involved in the maturation of pre-ribosomal RNA interacting with proteins like BOP1 and WDR12. This interaction signifies its role in the PeBoW complex important for proper cell division and growth. Pescadillo influences the cell cycle and has implications in regulating p53-dependent pathways which are critical for maintaining genomic stability.

Pathways

Pescadillo plays a part in the ribosome biogenesis pathway and p53 signaling pathway. In ribosome biogenesis it works alongside other proteins such as NPM1 and RPL5 to ensure accurate ribosome assembly necessary for protein synthesis. Within the p53 signaling pathway Pescadillo modulates cell cycle checkpoints preventing DNA damage from leading to cancerous growths through its modulation of p53 an important tumor suppressor.

Pescadillo's aberrant function or expression links to cancer and developmental disorders. Its overexpression commonly associates with breast cancer where it may serve as a biomarker due to its involvement in increased cell proliferation. Additionally mutations in genes regulating Pescadillo activity can lead to disorders in embryonic development as protein partners like RPL5 are implicated in these processes. Understanding its regulatory mechanisms offers potential therapeutic targets in oncology and developmental biology.
