Activity summary

The Pet1 protein also known as FEVER or Fev is a transcription factor with a mass of approximately 27 kDa. It is a member of the ETS (E26 transformation-specific) family and is principally expressed in serotonergic neurons of the raphe nuclei within the brain. This expression pattern is consistent with its role in the development and maintenance of specific neuronal subtypes.

Biological function summary

Pet1 drives the differentiation development and function of serotonergic neurons. It acts as a critical regulator by binding to DNA sequences to control the transcription of genes involved in neurotransmitter synthesis and transport. Pet1 forms complexes with other transcription factors enhancing its regulatory actions and it is especially important for the production of the neurotransmitter serotonin.

Pathways

Pet1 holds a significant role in the serotonin biosynthesis pathway. It regulates genes such as tryptophan hydroxylase 2 (TPH2) and the serotonin transporter (SERT) which are necessary for serotonin metabolism and its transport. Pet1 interacts with other proteins like GATA3 to fine-tune the serotonergic lineage specification and functions in a network that impacts mood regulation.

Associated diseases and disorders

Aberrations in Pet1 expression or function have links to conditions like depression and anxiety disorders. The dysfunction of serotonergic neurons in which Pet1 plays an important role contributes to these mental health disorders. Moreover alterations in Pet1 can influence the expression of receptor protein 5-HT1A which has implications in both the onset and treatment responses of depressive disorders.

    SH-SY5Y whole cell and nuclear extracts (30 μg) were separated by 12% SDS-PAGE, and the membrane was blotted with Pet1 antibody [HL2043] (ab308220) diluted at 1:1000. The HRP-conjugated anti-rabbit IgG antibody was used to detect the primary antibody.
