Paraffin-embedded human adrenal gland tissue stained for PEX13 using ab235043 at 1/200 dilution in immunohistochemical analysis.

After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized tissue using an HRP conjugated SP system.

PEX13 also called peroxin-13 is a protein involved in peroxisomal biogenesis. This protein has a molecular mass of approximately 42 kDa. PEX13 is expressed mainly in tissues that have a significant peroxisomal content such as the liver and kidney. It forms part of the peroxisome membrane where it facilitates the import of proteins into the peroxisome matrix. This process is essential for maintaining proper peroxisome function which plays a role in lipid metabolism and reactive oxygen species detoxification.
Biological function summary

PEX13 participates in the assembly of the peroxisomal importomer complex a multi-protein structure essential for peroxisomal matrix protein import. PEX13 interacts with other peroxins such as PEX14 and PEX5 to mediate the docking and translocation of cargo proteins that bear peroxisomal targeting signals. This process ensures that proteins required for peroxisomal functions are correctly imported impacting cellular homeostasis.

Pathways

PEX13 plays a role in peroxisome biogenesis and functioning pathways. In the peroxisome biogenesis pathway PEX13 directly interacts with other peroxins like PEX14 to assist in the formation and maintenance of functional peroxisomes. Alongside its interaction with PEX5 PEX13 is important in pathways facilitating the import of enzymes necessary for fatty acid beta-oxidation and hydrogen peroxide detoxification within peroxisomes.

PEX13 mutations are linked to Zellweger spectrum disorders a group of peroxisome biogenesis disorders. These conditions impair normal peroxisomal functions leading to severe developmental and neurological defects. Additionally some cases of neonatal adrenoleukodystrophy have associations with PEX13 dysfunction highlighting its role in peroxisome-related metabolic pathways. The disruption in PEX13 function in these disorders often involves its interactions with proteins such as PEX14 and PEX5 leading to improper protein import and peroxisomal dysfunction.

Component of the PEX13-PEX14 docking complex, a translocon channel that specifically mediates the import of peroxisomal cargo proteins bound to PEX5 receptor (PubMed : 28765278, PubMed : 8858165, PubMed : 9653144). The PEX13-PEX14 docking complex forms a large import pore which can be opened to a diameter of about 9 nm (By similarity). Mechanistically, PEX5 receptor along with cargo proteins associates with the PEX14 subunit of the PEX13-PEX14 docking complex in the cytosol, leading to the insertion of the receptor into the organelle membrane with the concomitant translocation of the cargo into the peroxisome matrix (PubMed : 28765278, PubMed : 8858165, PubMed : 9653144).
