PEX14 also known as Peroxisomal Membrane Protein 14 is an essential component of the peroxisomal membrane acting as an integral membrane protein. PEX14 has a mass of approximately 47 kDa and is expressed in many tissues including liver and kidney. It functions mechanically as a docking factor within the peroxisomal matrix enabling the import of proteins into peroxisomes which are cellular organelles responsible for various metabolic functions.
Biological function summary

PEX14 plays a significant role in maintaining peroxisome structure and function. It is part of the peroxisome importomer complex which facilitates the import of matrix proteins by interacting with peroxins such as PEX5. PEX14's interaction with these proteins ensures the proper translocation and sorting of enzymes critical for peroxisomal activities like fatty acid oxidation and reactive oxygen species detoxification.

Pathways

PEX14 is involved in key metabolic and cellular pathways including peroxisomal fatty acid beta-oxidation and ether lipid metabolism. It directly associates with PEX5 a peroxin that transports cargo proteins. In these pathways PEX14 plays a central role in maintaining metabolic processes that are essential for lipid metabolism and cellular detoxification.

PEX14’s malfunction or absence can contribute to Peroxisome Biogenesis Disorders (PBDs) such as Zellweger syndrome. These disorders significantly affect cellular metabolism and result in severe developmental issues. PEX14's interactions with proteins like PEX19 affect peroxisomal membrane biogenesis further linking PEX14 to these metabolic disorders and highlighting its importance in peroxisomal function.

Target data

Component of the PEX13-PEX14 docking complex, a translocon channel that specifically mediates the import of peroxisomal cargo proteins bound to PEX5 receptor (PubMed : 24235149, PubMed : 28765278, PubMed : 9653144). The PEX13-PEX14 docking complex forms a large import pore which can be opened to a diameter of about 9 nm (By similarity). Mechanistically, PEX5 receptor along with cargo proteins associates with the PEX14 subunit of the PEX13-PEX14 docking complex in the cytosol, leading to the insertion of the receptor into the organelle membrane with the concomitant translocation of the cargo into the peroxisome matrix (PubMed : 24235149, PubMed : 28765278). Plays a key role for peroxisome movement through a direct interaction with tubulin (PubMed : 21525035).
