PEX7 also known as Peroxisomal Biogenesis Factor 7 functions as a receptor for peroxisomal targeting signal 2 (PTS2). PEX7 has a molecular mass of approximately 39 kDa and localizes primarily in the cytoplasm and peroxisomes of cells. It is abundantly expressed in a variety of tissues including the liver kidney and heart. The expression level suggests that PEX7 plays an important mechanical role in peroxisomal import processes.
Biological function summary

PEX7 facilitates the import of peroxisomal matrix proteins with PTS2 signals by forming a complex with them. This complex guides the proteins to the peroxisome important for maintaining peroxisomal function and integrity. PEX7 interacts with other peroxisomal biogenesis factors such as PEX5 and PEX14 during this process. Proper functioning of PEX7 ensures that enzymes needed for lipid metabolism and other peroxisomal activities are correctly imported to their destination.

Pathways

Peroxisomal protein import involves PEX7 which integrates into broader systems like the peroxisome biogenesis and lipid metabolism pathways. In these pathways PEX7 associates with PEX5 an important player in both pathways facilitating the import of important enzymes for beta-oxidation of fatty acids. Without the proper coordination of these proteins the pathways would falter leading to metabolic irregularities.

Malfunction of PEX7 correlates strongly with Rhizomelic Chondrodysplasia Punctata (RCDP) and Zellweger Spectrum Disorders (ZSD). In these conditions PEX7's interaction with proteins like PEX5 becomes compromised leading to impaired peroxisomal protein import and subsequent cellular dysfunction. The dysfunctions highlight the importance of PEX7 in maintaining normal cellular physiology and highlight its relevance in therapeutic research targeting these diseases.
