Phosphofructokinase/fructose-26-bisphosphatase 2 often called PFKFB2 plays a significant role in the regulation of glycolysis and gluconeogenesis. This bifunctional enzyme has a molecular mass of approximately 55 kDa. PFKFB2 catalyzes the conversion between fructose-6-phosphate and fructose-26-bisphosphate. It is mainly expressed in cardiac tissue though expression has been noticed in other tissues such as skeletal muscle and liver.
PFKFB2 regulates the levels of fructose-26-bisphosphate an important metabolic signal molecule. This molecule modulates the activity of key enzymes in glycolysis. PFKFB2 forms part of a complex that controls the glycolytic flux in response to hormonal and nutritional changes. By increasing levels of fructose-26-bisphosphate it strongly activates phosphofructokinase-1 (PFK-1) pushing cells towards glycolytic metabolism under conditions of increased energy demand.
PFKFB2 directly influences the glycolysis and gluconeogenesis pathways. The regulation of these pathways by PFKFB2 ties it closely to energy metabolism. Additionally it has functional connections with enzymes such as phosphofructokinase-1 (PFK-1) and fructose-16-bisphosphatase (FBPase) helping to balance the glycolytic and gluconeogenic flux according to cellular needs.
Studies implicate PFKFB2 in conditions such as ischemic heart disease and certain cancers. Ischemic heart disease involves energy metabolism disruption where PFKFB2 plays a role in sustaining cardiac energy supply under oxygen-limited conditions. In cancer PFKFB2's altered expression correlates with the Warburg effect where cancer cells favor glycolysis even in the presence of oxygen. This connection makes PFKFB2 relevant to research fields exploring metabolic interventions in cancer therapeutics.
