The target PGAP3 also called post-GPI attachment to proteins 3 is a protein involved in the processing of glycosylphosphatidylinositol (GPI) anchors. It is encoded by the PGAP3 gene and has a molecular mass of around 104 kDa. This protein expresses highly in various tissues especially in the brain and testis. PGAP3 plays an important role in the correct remodeling of GPI anchors which are critical for the stable attachment of proteins to the cell membrane.
Biological function summary

PGAP3 removes unsaturated fatty acids from GPI anchors' lipid tails allowing the addition of saturated fatty acids. This process is essential for the function and stability of GPI-anchored proteins. Although not a member of a large protein complex PGAP3 works closely with other enzymes involved in GPI-anchor maturation. The remodeling of GPI anchors by PGAP3 ensures that proteins can adhere properly to the cell membrane and perform their functions.

Pathways

The correct localization of GPI-anchored proteins depends heavily on PGAP3 function. The protein plays an important role in the GPI-anchor biosynthesis pathway and changes in PGAP3 activity affect this pathway significantly. The remodeling of GPI anchors connects PGAP3 to proteins like PIG-N which also participates in this biosynthesis process. By ensuring the correct GPI anchor assembly PGAP3 helps maintain cellular communication and signaling pathways.

Alterations in PGAP3 function associate with conditions such as intellectual disability and GPI-anchor deficiency syndrome. Mutations in the PGAP3 gene disrupt normal GPI-anchor maturation leading to a range of developmental and neurological abnormalities. The protein is also linked with PIGT another enzyme involved in the GPI biosynthesis pathway implicating these proteins in the onset of specific disorders when disrupted. Understanding the role of PGAP3 in these diseases aids in developing potential targeted therapies.

Required for phospholipase A2 activity that removes an acyl-chain at the sn-2 position of GPI-anchors during the remodeling of GPI.
