Supplementary information

PGC1 beta also known as PPARGC1B or PERC is an important transcriptional coactivator involved in the regulation of genes related to energy metabolism. It has a molecular weight of approximately 102 kDa. Researchers have found PGC1 beta expressed in various tissues with high metabolic activity including heart skeletal muscle liver and brown adipose tissue. By interacting with numerous nuclear receptors and other transcription factors PGC1 beta modulates the transcription of genes important for mitochondrial biogenesis and function.
Biological function summary

PGC1 beta plays a significant role in the regulation of energy homeostasis and adaptive thermogenesis. It partners with specific DNA-binding proteins as part of large coactivator complexes to enhance transcriptional activity. Essential to maintaining energy balance PGC1 beta facilitates the switch between glucose and fatty acid oxidation in response to metabolic demands. This coactivator's ability to interact with key metabolic regulators such as PPARs and ERRα highlights its vital role in cellular energy regulation.

Pathways

PGC1 beta has an important role in energy metabolism including the oxidative phosphorylation and fatty acid oxidation pathways. It functions closely with PPAR-γ and PPAR-α as well as other proteins like NRF1 and NRF2 to regulate the expression of genes involved in mitochondrial function. PGC1 beta activates these pathways contributing to efficient energy utilization and mitochondrial biogenesis important for adapting to varying energy requirements.

PGC1 beta has associations with metabolic diseases like type 2 diabetes and obesity. Altered expression or function of PGC1 beta can disrupt normal metabolic regulation leading to energy imbalance and insulin resistance. Studies link PGC1 beta modulation to liver disease as well connecting it with proteins such as SIRT1 which also plays a role in metabolic homeostasis and cellular stress responses. Understanding PGC1 beta's involvement in these diseases could provide insights into potential therapeutic approaches for metabolic disorders.

Target data

Plays a role of stimulator of transcription factors and nuclear receptors activities. Activates transcriptional activity of estrogen receptor alpha, nuclear respiratory factor 1 (NRF1) and glucocorticoid receptor in the presence of glucocorticoids. May play a role in constitutive non-adrenergic-mediated mitochondrial biogenesis as suggested by increased basal oxygen consumption and mitochondrial number when overexpressed. May be involved in fat oxidation and non-oxidative glucose metabolism and in the regulation of energy expenditure. Induces the expression of PERM1 in the skeletal muscle in an ESRRA-dependent manner.
