Phosphoglycerate kinase 2 (PGK2) also known as testis-specific phosphoglycerate kinase is an enzyme with a molecular mass of approximately 45 kDa. It functions in glycolysis by catalyzing the reversible conversion of 13-bisphosphoglycerate to 3-phosphoglycerate producing ATP in the process. PGK2 expression is restricted mostly to the testes where it plays an important role in sperm development and motility. Unlike its isozyme counterpart PGK1 which is expressed ubiquitously PGK2 is testis-specific.
Biological function summary

The enzyme participates in the important processes of energy production and spermatozoa development. While PGK2 does not form part of any multi-protein complex it is an integral component of the glycolytic pathway contributing to ATP production necessary for sperm motility. Its expression is regulated during spermatogenesis highlighting its role in male fertility.

Pathways

PGK2 operates within the glycolytic pathway a major metabolic route for energy production. Along with glyceraldehyde-3-phosphate dehydrogenase PGK2 catalyzes important reactions that facilitate the conversion of glucose to pyruvate yielding high-energy ATP molecules. By functioning in tandem with other glycolytic enzymes PGK2 ensures the efficient production of energy needed for cellular functions especially during spermatogenesis.

Defects or alterations in PGK2 have associations with male infertility notably due to impaired sperm function. Disorders like spermatogenic failure and related conditions arise when PGK2 activity diminishes. Its enzyme partner glyceraldehyde-3-phosphate dehydrogenase-S is also linked to sperm motility issues. Understanding PGK2’s role aids in identifying therapeutic targets to address such reproductive challenges.
