PHF10 was immunoprecipitated from 0.35 mg HeLa (human cervix adenocarcinoma epithelial cell) whole cell lysate 10 ug with ab259946 at 1/30 dilution (2ug in 0.35mg lysates). Western blot was performed on the immunoprecipitate using ab259946 at 1/1000 dilution. VeriBlot for IP secondary antibody(HRP)(ab131366) was used at 1/5000 dilution.

Researchers have identified PHF10 as a component of the BAF (BRG1-associated factor) chromatin remodeling complex. It is widely expressed in various tissues including the brain where it contributes to neural development and function. Its expression can have tissue-specific variations that reflect its diverse roles in different cellular contexts.
Biological function summary

PHD finger protein 10 participates in the regulation of gene transcription and chromatin architecture. It functions as part of the BAF complex which orchestrates the dynamic remodeling of chromatin to either activate or repress specific genes. These pathways play fundamental roles in ensuring proper development and maintaining cellular homeostasis where PHF10 activity impacts both normal cellular function and response to environmental changes.

Dysregulation of PHF10 function associates with neurodevelopmental disorders and certain forms of cancer. Altered PHF10 expression or mutations can disrupt normal chromatin remodeling activities leading to improper gene expression profiles. In the context of neurodevelopmental disorders PHF10 misfunction can impact genes critical for nervous system development. In some cancers abnormal interactions between PHF10 and proteins like SMARCC1 another BAF complex member can result in the derangement of gene regulatory networks therefore promoting oncogenic processes.

Involved in transcription activity regulation by chromatin remodeling. Belongs to the neural progenitors-specific chromatin remodeling complex (npBAF complex) and is required for the proliferation of neural progenitors. During neural development a switch from a stem/progenitor to a post-mitotic chromatin remodeling mechanism occurs as neurons exit the cell cycle and become committed to their adult state. The transition from proliferating neural stem/progenitor cells to post-mitotic neurons requires a switch in subunit composition of the npBAF and nBAF complexes. As neural progenitors exit mitosis and differentiate into neurons, npBAF complexes which contain ACTL6A/BAF53A and PHF10/BAF45A, are exchanged for homologous alternative ACTL6B/BAF53B and DPF1/BAF45B or DPF3/BAF45C subunits in neuron-specific complexes (nBAF). The npBAF complex is essential for the self-renewal/proliferative capacity of the multipotent neural stem cells. The nBAF complex along with CREST plays a role regulating the activity of genes essential for dendrite growth (By similarity).
