Supplementary information

This supplementary information is collated from multiple sources and compiled automatically.

PHF16 also known as JADE3 is a protein with a mass of approximately 90 kDa. It is a member of the plant homeodomain finger family of proteins. PHF16 contains domains that mediate interactions with other proteins indicating its role in chromatin remodeling and transcription regulation. This protein is widely expressed in tissues including brain liver and lung. Its localization in these tissues suggests its involvement in various cellular processes.
Biological function summary

PHF16 interacts with a range of molecular complexes related to gene expression regulation. It is a component of the HBO1 complex which plays a role in histone acetylation. This complex modifies chromatin structure to facilitate access to DNA for transcription machinery. PHF16 acts by binding to specific histone marks participating in the regulation of transcriptional activation of genes. These functions make it an important player in maintaining proper gene expression levels across diverse cell types.

Pathways

PHF16 influences transcriptional regulation pathways and epigenetic modification pathways. Within these frameworks it closely interacts with other proteins like HBO1 and ING5. PHF16 contributes to the modification of histone acetylation patterns which is important for transcriptional regulation. It assists in setting the epigenetic landscape that determines the transcriptional output of genes involved in cell cycle and development.

Altered expression or mutation of PHF16 links to several pathological conditions including cancer and developmental disorders. For example dysregulation of PHF16 expression may contribute to tumorigenesis particularly in breast cancer due to its role in regulating genes essential for cell proliferation. Moreover interactions with proteins like HBO1 and ING5 highlight its importance in pathways disrupted in developmental diseases where epigenetic misregulation plays a significant role. Understanding PHF16's function and interactions provides critical insights into its potential as a therapeutic target in these disorders.
