The PHLDA1 protein also known as T-cell death-associated gene 51 (TDAG51) weighs approximately 34 kDa. This protein plays roles in regulating apoptosis and cell proliferation. It expresses mainly in the heart kidney pancreas and placenta. PHLDA1 consists of a pleckstrin homology domain which mediates its binding to phosphoinositides allowing it to interact with cell membranes and influence cellular signaling pathways.
Biological function summary

PHLDA1 contributes to cellular processes such as apoptosis survival and proliferation. It does not appear to be part of a larger complex but engages in interactions that regulate cell fate. PHLDA1 mediates cellular response to stress and participates in cellular growth regulation by modulating the activity of other signaling molecules. This function makes it important in maintaining cellular homeostasis.

Pathways

Researchers connect PHLDA1 to the Akt signaling pathway and the TNF receptor pathway. In the Akt signaling pathway PHLDA1 interacts with proteins such as Forkhead box O3 (FOXO3) to influence cell survival and apoptosis. In the TNF receptor pathway PHLDA1 participates in modulating the balance between cell survival and programmed cell death impacting inflammatory responses.

PHLDA1 relates to cancer and neurodegenerative diseases. Its expression levels alter in certain types of cancers such as breast and prostate cancer where it may have either tumor-suppressing or oncogenic roles depending on context. In Alzheimer's disease connections exist between PHLDA1 dysregulation and proteins like amyloid precursor protein (APP) suggesting a role in disease progression. Understanding PHLDA1’s role in these conditions may offer insight into new therapeutic strategies.
