An ELISA plate was coated with crude extract of B. cereus (100 μL, 1 μg/mL). After blocking with 5% skimmed milk, 100 μL of antibody at the indicated dilutions was added to each well. HRP-conjugated goat anti-mouse IgM (100 μL, X2000 dilution) was added.

Phospholipase C also known as PLC is an enzyme that catalyzes the hydrolysis of phosphatidylinositol 45-bisphosphate (PIP2) into diacylglycerol (DAG) and inositol trisphosphate (IP3). It has several isoforms such as PLC beta and PLC gamma each with different molecular sizes. For example PLC gamma typically has a mass of about 130 kDa. This enzyme is expressed in many tissues including the brain liver and pancreas where it plays an important role in cellular signaling.
Biological function summary

The activity of Phospholipase C generates second messengers that are important for intracellular signal transduction. It functions as part of larger signaling complexes and can be involved in regulating various cellular processes like proliferation differentiation and apoptosis. This role makes it an important regulator in many physiological responses. The enzyme often interacts with membrane-bound receptors linking extracellular signals to internal cellular responses.

Pathways

Phospholipase C is an important player in the phosphatidylinositol signaling pathway and is interconnected with the calcium signaling pathway. Within these pathways PLC activity triggers the release of calcium from internal stores which in turn can activate calcium-dependent proteins such as protein kinase C. These interactions aid in modulating cellular responses to external stimuli.

PLC abnormalities have been linked to conditions like cancer and bipolar disorder. Altered PLC function can disrupt normal cellular signaling contributing to the pathogenesis of these diseases. In cancer abnormal PLC activity is tied to uncontrolled cell growth often interacting with proteins like Ras to drive oncogenic processes. In bipolar disorder disruptions in PLC function affect neurotransmitter pathways implicating changes in proteins associated with synaptic signaling.
