- Flow Cyt (Intra)
Unknown
Flow Cytometry (Intracellular) - Anti-PHPT1 antibody [001] - BSA and Azide free (AB276060)
Intracellular flow cytometric analysis of Human PHPT1 expression on THP1 cells. The cells were treated according to manufacturers manuall, stained with ab276060, then a FITC-conjugated secondary antibody (black line) compared with a negative control (grey line). The Fluorescence histograms were derived from gated events with the forward and side light-scatter characteristics of intact cells.
- IHC-P
Unknown
Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-PHPT1 antibody [001] - BSA and Azide free (AB276060)
Immunochemical staining of human PHPT1 in human skeletal muscle with ab276060 at 1/5000 dilution, formalin-fixed paraffin embedded sections.
- ICC
Unknown
Immunocytochemistry - Anti-PHPT1 antibody [001] - BSA and Azide free (AB276060)
Immunofluorescence staining of Human PHPT1 in HeLa cells. Cells were fixed with 4% PFA, permeabilzed with 0.3% Triton X-100 in PBS, blocked with 10% serum, and incubated with ab276060 at 1/60 dilution at 37°C 1 hour. Then cells were stained with the Alexa Fluor® 488-conjugated Goat Anti-rabbit IgG secondary antibody (green) and counterstained with DAPI (blue). Positive staining was localized to cytoplasm.

Biological function summary
This protein modulates signal transduction pathways by dephosphorylating proteins which can affect cellular signals and functions. Although not part of a larger protein complex PHPT1 acts by influencing histidine phosphorylation status therefore impacting cellular communication. The regulation of phosphorylation states by PHPT1 affects processes like growth differentiation and metabolism underlining its significance in cellular functions.
Pathways
PHPT1 engages in histidine phosphorylation pathways a lesser-known aspect compared to serine threonine and tyrosine phosphorylations. It plays a significant role in cellular signaling networks where related proteins include kinases that add phosphate groups to histidine residues. By modifying the phosphorylation state of certain proteins PHPT1 influences important pathways such as those involving signaling cascades that regulate cell growth and apoptosis.
