PHRF1 also known as PHRF1 actin binding protein functions as an E3 ubiquitin ligase within cells. It has a molecular mass of approximately 91 kDa and is widely expressed across various tissues with higher expression levels observed in the nucleus. PHRF1 interacts with and modifies other proteins through ubiquitination which signals them for degradation or alters their activity within the cell.
This protein influences cellular processes by interacting with transcriptional co-repressors and partners in transcriptional repression. PHRF1 plays a significant role in regulating gene expression often forming complexes with other proteins involved in chromatin remodeling and transcriptional regulation. Its activity helps maintain proper gene expression levels which is critical for cellular functions and development.
PHRF1 participates in key biological pathways such as the TGF-beta signaling pathway. It collaborates with SMAD proteins which are important in the TGF-beta signaling cascade influencing cell growth and differentiation. Another important connection is with the Hippo signaling pathway where PHRF1 interacts with proteins involved in controlling organ size and tissue homeostasis.
Mutations or misregulation of PHRF1 have been linked to cancer development particularly breast and lung cancers. Its role in regulating gene expression and cellular growth connects PHRF1 to tumor suppression mechanisms. Additionally it interacts with proteins like HIC1 and CHD8 which modulate tumorigenic pathways and might have implications in cancer progression.
