Binds to activated (phosphorylated) protein-Tyr kinases, through its SH2 domain, and acts as an adapter, mediating the association of the p110 catalytic unit to the plasma membrane. Necessary for the insulin-stimulated increase in glucose uptake and glycogen synthesis in insulin-sensitive tissues. Plays an important role in signaling in response to FGFR1, FGFR2, FGFR3, FGFR4, KITLG/SCF, KIT, PDGFRA and PDGFRB. Likewise, plays a role in ITGB2 signaling (PubMed:17626883, PubMed:19805105, PubMed:7518429). Modulates the cellular response to ER stress by promoting nuclear translocation of XBP1 isoform 2 in a ER stress- and/or insulin-dependent manner during metabolic overloading in the liver and hence plays a role in glucose tolerance improvement (PubMed:20348923).

Activity summary

The PI 3 Kinase p85 alpha + Gamma also known as PI3K is a regulatory subunit of the class IA phosphoinositide 3-kinases. PI3K is comprised of the p85 alpha (approximately 83 kDa) and p110 gamma subunits among others and plays an important role in cell signaling. The p85 alpha subunit is primarily expressed in immune cells liver and muscles while the p110 gamma is mostly found in leukocytes. This kinase is deeply involved in a variety of cellular processes like cell growth proliferation differentiation motility and survival.

Biological function summary

The PI 3 Kinase p85 alpha + Gamma is an important regulator in signal transduction pathways and forms a heterodimer complex with p110 catalytic subunits. This kinase complex is activated by various growth factors and hormones through receptor tyrosine kinases. Upon activation PI3K phosphorylates phosphatidylinositol (45)-bisphosphate (PIP2) to produce phosphatidylinositol (345)-trisphosphate (PIP3) an essential second messenger that recruits downstream signaling proteins like AKT to the plasma membrane. This signal cascade regulates numerous cellular functions affecting homeostasis and adaptive responses.

Pathways

The PI 3 Kinase p85 alpha + Gamma plays a significant role in the PI3K/AKT/mTOR and MAPK signaling pathways. These pathways are essential in controlling cell metabolism survival and growth. Within these cascades PI3K phosphorylates PIP2 to PIP3 creating a docking site for AKT which subsequently becomes activated and relays signals to downstream effectors including mTOR and GSK-3β. The proper functioning of these pathways is necessary for maintaining cellular equilibrium and their dysregulation might lead to pathological conditions.

Associated diseases and disorders

Aberrant PI 3 Kinase p85 alpha + Gamma activity is implicated in cancer and immune disorders. In particular its malfunction is notable in various cancers including breast and prostate cancer where mutations can lead to constitutive activation of PI3K signaling promoting tumorigenic processes. Additionally mutations or altered expression of PI3K can contribute to immune-related conditions by affecting immune cell function and survival. Interactions between PI3K and other proteins such as PTEN a tumor suppressor represent key nodes in understanding these disease mechanisms and offer potential therapeutic targets.

    ab74136 at 1/200 dilution staining PI 3 Kinase p85 alpha + Gamma in mouse embryonic stem cells by Immunocytochemistry/ Immunofluorescence. Cells were fixed in paraformaldehyde, permeabilized in 1:1 methanol, acetone and blocked in 1% serum for 30 minutes at 25°C. The primary antibody was used at 1/100 dilution in Goat block dillution buffer and incubated with sample at 25°C for 1 hours. An Alexa Fluor® 488 conjugated Goat polyclonal to rabbit IgG (H+L) was used as secondary at 1/500 dilution.
